Inhibitory Effect of Scoparone on U46619-Induced Platelet Aggregation via Regulation of PI3K/Akt and MAPKs

Abstract
Damage to blood vessels results in a rapid hemostatic response in order to minimize blood loss and maintain normal circulation. Platelet activation and aggregation are essential for this process. However, excessive or abnormal platelet aggregation may result in cardiovascular diseases such as thrombosis, stroke, and atherosclerosis. Therefore, it is important to discover a substance capable of regulating platelet activation, and suppressing the aggregation reaction for the prevention and treatment of cardiovascular diseases. Scoparone, found mainly in the roots of Artemisia or Scopolia plants, is reported to exert pharmacological effects on immunosuppression and vasodilation, but studies on platelet aggregation and its mechanisms are lacking. This study, therefore, undertook to determine the effects of scoparone on U46619-induced human platelet aggregation, and on the major regulation of TXA2 production and intracellular granule secretion (ATP and serotonin release). In addition, we examined the effects of scoparone on the phosphorylation of phosphoproteins PI3K/Akt and MAPKs, which are involved in signal transduction in platelet aggregation. We observed that scoparone significantly inhibits the phosphorylation of PI3K/Akt and MAPKs, which subsequently and significantly inhibit platelet aggregation through TXA2 production and intracellular granule secretion (ATP and serotonin release). We, therefore, propose that scoparone is an antiplatelet substance that regulates phosphorylation of phosphoproteins such as PI3K/Akt and MAPKs, and has the potential to be a valuable preventive and therapeutic agent for platelet-derived cardiovascular diseases.
